Science Research sophomores bring advanced DNA analysis to Ursuline

by Shannon Dufresne

In December, Ursuline sophomores enrolled in Science Research transformed a classroom into a working research lab, completing a complex DNA barcoding experiment entirely on campus. Guided by Science teacher Dr. Oxana Litvine, an industry professional, students used professional-grade tools and protocols to generate data that contribute to an authentic scientific database and real-world research outcomes. The lab marked the first time students conducted this advanced work at Ursuline, rather than making the day-long trip to Cold Spring Harbor Laboratory.

Students began work early in Room 309 of Ursuline's North Building, spending a full school day immersed in the project. While the change in setting presented challenges, it also highlighted the depth and flexibility of Ursuline's Science Research program, providing students with hands-on experience in authentic scientific inquiry while strengthening their problem-solving skills.

DNA barcoding is a signature component of the sophomore Science Research curriculum and Ursuline's Citizen Science Club. The project introduces students to the full research process, from data collection to analysis. For many sophomores, it serves as a valuable first step toward their first complete, competition-ready research project.

This year, five sophomore projects are being prepared for submission to the Regeneron WESEF (Westchester Science & Engineering Fair). Three projects were already completed through Citizen Science, while two were finalized during the December lab, with the entire class participating, giving some students a second opportunity to experiment.

Conducting the lab on campus required students to use professional-level techniques. Working in teams, students extracted DNA from a wide range of specimens, including plants collected on Ursuline’s campus and invertebrates gathered during the annual Marine Biology trip to Long Island Sound. Each specimen required different methods and primers, demanding care planning and collaboration.

Susanna Connelly '28, who previously completed the experiment in May, welcomed the challenge. "It took some creativity to get everything how we wanted it to be, and it was a really great learning opportunity,” she said. The experience left her confident in her ability to guide others through the process.

Dr. Litvine emphasized that the project's primary purpose is to support students’ interests and provide meaningful experiences that enhance their college applications. By bringing advanced research opportunities to campus, Ursuline increases access for students balancing academics, athletics, and extracurricular commitments while still offering college-level scientific experiences.

Connelly describes the program as "valuable preparation for future lab work," noting that it builds confidence. "It's a really cool opportunity! The DNA barcoding project is like a jumping-off point for me," she said. Students graduate from the program with a strong foundation in laboratory techniques, research communication, and independent inquiry skills valued across disciplines.

As competition season approaches, momentum builds. During the 2024–25 academic year, Science Research students earned 15 competition awards with just 27 participants, reflecting both the rigor of the program and the individualized support students receive.

Through experiences like DNA barcoding, Ursuline's Science Research program offers students opportunities rarely found at the high school level, preparing them to think critically, work collaboratively, and pursue ambitious questions with confidence.
